New inhibitors of elusive enzymes promise to be valuable scientific tools

ScienceDaily (Oct. 31, 2012) ? Scientists at The Scripps Research Institute (TSRI) have discovered the first selective inhibitors of an important set of enzymes. The new inhibitors, and chemical probes based on them, now can be used to study the functions of enzymes known as diacylglycerol lipases (DAGL), their products, and the pathways they regulate. Early tests in mouse macrophages suggest that DAGL-inhibiting compounds might also have therapeutic uses, for they suppress the production of a pro-inflammatory molecule that has been implicated in rheumatoid arthritis and related conditions.

"We've developed the first set of chemical probes that effectively allows one to study these DAGL enzymes in living cell and animal models," said Benjamin F. Cravatt, chairman of the Department of Chemical Physiology, professor in the Dorris Neuroscience Center and member of the Skaggs Institute for Chemical Biology at TSRI. Cravatt and his laboratory conducted the new study, published in the current issue of the journal Nature Chemical Biology.

Important But Poorly Understood

DAGL enzymes have been of interest mainly because of their role in making 2-AG (2-Arachidonoylglycerol), an important cannabinoid that is naturally produced in humans and other mammals. Cannabinoids are named for Cannabis (marijuana) plants, because they stimulate the same cellular receptors that are hit by marijuana's active ingredients. Drugs that can enhance 2-AG's signaling in the nervous system are being developed as treatments for pain, depression and anxiety.

But 2-AG exists in various tissues throughout the body, and on the whole, its functions are not well understood. Until now researchers have lacked enzyme inhibitors that can usefully probe those functions by selectively shutting off 2-AG's production. "Existing DAGL inhibitors block many other enzymes, are not very potent, and do a poor job of getting into cells," Cravatt said. "There has been a need for better chemical tools in this area."

Cravatt's laboratory had previously developed a set of compounds that act as potent inhibitors of serine hydrolases -- the broad enzyme family to which DAGL enzymes belong. In the new study, Cravatt's team, including first author Ken Hsu, a Hewitt Foundation postdoctoral researcher in the Cravatt laboratory, screened a library of these compounds for specific activity as DAGL inhibitors.

A Big Improvement

After finding a promising lead compound, Hsu and his colleagues chemically optimized it to obtain KT109 and KT172. The former selectively inhibits DAGL?, the main enzymatic producer of 2-AG outside the nervous system. KT172 inhibits both DAGL? and DAGL?, which is principally responsible for making 2-AG within the nervous system.

In a big improvement over previously described DAGL inhibitors, KT109 and KT172 are highly selective (i.e., they do not block many other, non-DAGL enzymes) and active in cells and animals. By analyzing the structures of their initial DAGL inhibitors, the team was also able to devise a new DAGL-tailored activity-based probe that binds to the active site of DAGLs and fluorescently labels these low-abundance and difficult-to-detect enzymes in cell or tissue samples. "Without the DAGL-specific probe, we would have found it very difficult to develop, optimize and confirm target engagement for our DAGL inhibitors," Hsu said.

In neuron-like mouse cells, human prostate cancer cells, and mouse liver cells and macrophages (a type of immune cell that is frequently involved in inflammatory conditions), the DAGL inhibitors were able to inactivate DAGL? activity. "At the optimal doses used, we were able to achieve selective and near-complete inhibition of the enzyme," said Hsu. In these cell and animal studies, the inhibitors also reduced levels of 2-AG as well as arachidonic acid, another bioactive lipid that DAGL enzymes can regulate.

New Questions

2-AG is known to have an anti-inflammatory effect when it activates cannabinoid receptors on macrophages. Thus, one might expect that knocking down 2-AG production with a DAGL inhibitor would have a pro-inflammatory effect. Instead, Hsu, Cravatt and their colleagues found that blocking DAGL in mouse macrophages that had been stimulated with pro-inflammatory agents markedly lowered their secretion of TNF?, a major inflammatory signaling molecule.

Blocking DAGL has potential effects on multiple lipid signaling pathways in cells, and the researchers aren't yet certain which of these effects explains the surprising suppression of TNF?. "The effect is dependent on DAGL?, though, because we see the same result in DAGL? knockout mice," said Hsu. Cravatt added that their observations of the unexpected DAGL-inhibition effects in mouse macrophages could be due to the suppression of pro-inflammatory eicosanoids that derive from downstream metabolites regulated by DAGL?.

TNF? is a key instigator of the inflammation seen in rheumatoid arthritis, and antibodies directed against TNF? are now front-line therapies for the condition. "What we've done so far is just early-stage cell biology, but conceivably the further optimization of our DAGL inhibitors could result in a new type of anti-inflammatory drug that also works against arthritis and related conditions," Cravatt said.

Cravatt and his team are now studying the pathways through which the new inhibitors have this anti-inflammatory effect. They also plan to develop new inhibitors that will selectively block DAGL? and 2-AG production in the central nervous system.

The other co-authors of the study, "DAGL? inhibition perturbs a lipid network involved in macrophage inflammatory responses," were Katsunori Tsuboi, Alexander Adibekian, Holly Pugh and Kim Masuda, all of the Department of Chemical Physiology at TSRI.

Stressed-Out Parents More Likely To Serve Up Fast FoodYour ...

Stressed-out parents were found to be more likely to serve their children fast food, and, perhaps not surprisingly, more likely to have obese kids. The study, published on Monday in the journal Pediatrics, looked at survey responses from 2,119 parents and caregivers of kids ranging in age from 3
to 17. The researchers then measured stress based on self-reported data from the respondents.

?Stress in parents may be an important risk factor for child obesity and related behaviors,? said Dr. Elizabeth Prout-Parks, a physician nutrition specialist at The Children?s Hospital of Philadelphia, who led the study. ?The severity and number of stressors are important.?

Some of the stressors weighing heavily on parents with obese children included poor physical and mental health, money woes, and being a single parent ? the last factor having the strongest link to child obesity. Although previous research has found a connection between parental stress and child obesity, the researchers say that their study covered a more diverse population, both ethnically and socioeconomically, than did previous studies.

Berlusconi sentenced to four years for tax evasion

MILAN (Reuters) - Former Italian Prime Minister Silvio Berlusconi was sentenced to four years in jail on Friday for tax fraud in connection with the purchase of broadcasting rights by his Mediaset television company.

The 76-year-old billionaire, who was convicted three times during the 1990s in the first degree before being cleared by higher courts, has the right to appeal the ruling two more times before the sentence becomes definitive. He will not be jailed unless he loses the final appeal.

The ruling comes two days after Berlusconi confirmed he would not run in next year's elections as the leader of his People of Freedom (PDL) party, ending almost 19 years as the dominant politician of the center-right.

Milan judge Edoardo d'Avossa told a packed court that between 2000 and 2003, there had been "a very significant amount of tax evasion" and "an incredible mechanism of fraud" in place around the buying and selling of broadcast rights.

The court's written ruling said Berlusconi showed a "natural capacity for crime".

Berlusconi lawyers Piero Longo and Niccolo Ghedini said the ruling was "totally divorced from all judicial logic", adding that they hoped the "atmosphere" at the appeals courts would be different.

Berlusconi, one of Italy's richest men, became prime minister for a second time in 2001 after winning a landslide election victory. Even while he was prime minister, he remained in effective charge of Mediaset even though he had handed over control of day-to-day operations, the court said.

The four-time prime minister and other Mediaset executives stood accused of inflating the price paid for TV rights via offshore companies controlled by Berlusconi and skimming off part of the money to create illegal slush funds.

The investigation focused on television and cinema rights that Berlusconi's holding company Fininvest bought via offshore companies from Hollywood studios.

The court also ordered damages provisionally set at 10 million euros ($13 million) to be paid by Berlusconi and his co-defendants to tax authorities.

"POLITICAL HOMICIDE"

The flamboyant Berlusconi, who is still on trial in a separate prostitution case, resigned as prime minister a year ago as Italy faced a Greek-style debt crisis, handing the reins of government to economics professor Mario Monti.

Angelino Alfano, secretary of the PDL, said the ruling proved once again "judicial persecution" of the media magnate, while political rival Antonio Di Pietro, a former magistrate, hailed the decision, saying "the truth has been exposed".

Should the ruling be confirmed on appeal, Berlusconi would also be forbidden from holding public office for five years, and from being a company executive for three years.

"This is not a sentence, but an attempt at political homicide," Fabrizio Chicchito, the PDL's chief whip in the Chamber of Deputies, said referring to the ban from holding office.

Now that Berlusconi has said he will pull out of politics, he may be focusing more on his business empire, which includes Mediaset, AC Milan soccer club, and Internet bank Mediolanum.

Shares in Mediaset, Italy's biggest private broadcaster, fell as much as 3 percent after the ruling, and are down about 50 percent in the last year.

The broadcaster has been struggling against rivals like News Corp's broadcaster Sky Italia and a host of online media, while its core advertising revenues are feeling the pinch of the recession.

The court acquitted Mediaset chairman and long-term Berlusconi friend Fedele Confalonieri, for whom prosecutors had sought a sentence of three years and four months.

Berlusconi has owned AC Milan since 1986 and they have been European champions five times under his leadership. But the club's fortunes have dipped in the past couple of seasons amid cost cutting, prompting repeated rumors of its possible sale.

He also is still on trial in the separate "Rubygate" case in which he is accused of paying for sex with a teenaged nightclub dancer when she was under 18 and thus too young to be paid legally as a prostitute. He denies the charges.

Cybersecurity chief: Attacks on US banks tough to pin down

Cybersecurity getting increased attention from US government after a wave of Internet attacks on US banks. Pentagon blames Iran, but cybersecurity expert Gil Shwed says they could come from any developed country.

The man often credited as being the father of Internet defense says it's still unclear where a recent wave of hacking attacks targeting the U.S. financial industry are coming from.

?Shwed said today "Iran is definitely capable of launching these kinds of attacks but so is just about any other developed country."

?In the last several weeks reports have surfaced that several major American banks have been hit by a wave of Internet-based attacks slowing down or temporarily cutting off service.? Officials at the Pentagon were the first to?point the finger at the government of Iran. Shwed agrees it is possible Iran is to blame but cautions it's very difficult to tell where attacks come from because they can be diverted over and over to cover up the actual source.

?Without admitting guilt, Iranian officials have boasted about their increasing capability to carry out such attacks.

?While the Islamic Republic of Iran is widely blamed, a group calling itself the Izz ad-Din al-Qassam Cyber Fighters Group has claimed responsibility. Their name comes from a Muslim leader that directed waves of murderous attacks targeting Jews living in the Middle East in the 1920s and 1930s.? The Izz ad-Din al-Qassam Cyber Fighters dubbed their internet attacks "Operation Ababil" (Ababil is Farsi for swallow ? the Iranian government also has a fleet of Unmanned Aerial Vehicles by the same name).

?In a statement filed on the internet late Tuesday night the group said, "Operation Ababil is retaliation in response of the organized insulting to the Prophet of Islam done by some arrogant western governments." The statement then again took credit for attacks on several American banks and threatened to carry out more.

On Wednesday, a New York Times report said Iranian hackers were also able to break into the?computer system of oil giant Saudi Aramco?earlier this year and wipe out almost three quarters of the data saved on the company's servers.

?There are signs the U.S. Government is starting to take these threats more seriously. Earlier this month Secretary of Defense Leon Panetta?sounded the alarm?on the nation's cyber-security saying "an aggressor nation or extremist group could gain control of critical switches and derail passenger trains, or trains loaded with lethal chemical. They could contaminate the water supply in major cities, or shut down the power grid across large parts of the country... a cyber-Pearl Harbor that would cause physical destruction and loss of life, paralyze and shock the nation, and create a profound new sense of vulnerability.''

Shwed echoed the Secretary of Defense's comments saying "the risk is absolutely there. But it isn't just government and infrastructure agencies that need to be on guard.? Every individual with a computer is at risk. These organizations, wherever they are, can infect tens of
thousands or even millions of computers."

Shwed added keeping your company or your individual computer or mobile device's security software updated is essential.

Despite the recent wave of cyber-attacks Check Point's stock has taken a hit in the last six months, it's down about 30 percent. Shwed blames the drop in large part to concerns over the European slowdown and the impact that will have on growth. But he optimistically says "the company is doing well whether profits increase 20 percent or decrease 20 percent." He also noted despite the recent downturn, shares are up almost 35 percent over the last three years.

Pending budget cuts will further jeopardize global leadership in research

WASHINGTON, DCOctober 25, 2012Biomedical and health research and development (R&D) spending from all sources declined by more than $4 billion or 3% between FY10 and FY11 according to Research!America's 2011 U.S. Investment in Health Research report. This represents the first drop in overall spending since Research!America began compiling the data in 2002.

The decline follows an uptick in research funding attributed to the American Recovery and Reinvestment Act (ARRA), which allocated $10.4 billion to the National Institutes of Health (NIH) over two fiscal years (2009-2010). The overall downward trend in R&D spending is coming at a time when other nations are ramping up their own investments in research, and meanwhile, pending across-the-board budget cuts (sequestration) could reduce federal biomedical and health research funding by 8%-10% or more.

"Insufficient funding, coupled with deep budget cuts under sequestration, could be devastating for research," said Research!America's chair, former Congressman John E. Porter. "Our global competitiveness hinges on a robust investment that will support bright scientific minds, create high-quality jobs and provide a catalyst for private sector innovation."

Research!America's 2011 U.S. Investment in Health Research report shows varying levels of health research funding in the private and public sector. For example, federal funding for research totaled $39.5 billion in FY11, a 14% decrease from the previous year's total of $45.9 billion. Agency funds were distributed across all 50 states to hospitals, universities, independent research institutes and small businesses. Under sequestration, the NIH would lose $2.53 billion in funding in FY13.

"As R&D spending abroad outpaces federal investments here at home, U.S. companies will set up shop in countries with stronger policies to support research," said Mary Woolley, president and CEO of Research!America. "We cannot afford to become complacent as cures for Alzheimer's, cancer, heart disease and other serious health threats remain a priority for every American."

Overall, private industry has continually increased investments in R&D a total of $77.6 billion in 2011, a 1.4% increase from 2010, despite inflationary pressure and the economic recession. The pharmaceutical industry increased its investment to $38.5 billion, a 3% increase from the previous year. In contrast, biotechnology investment declined by nearly $800 million, or 3%. The medical device and technology sector slightly increased investment in research, totaling $9.8 billion. Currently, more than 80% of R&D among PhRMA member companies is conducted in the United States, but R&D spending abroad has more than doubled over the past decade.

Aside from federal and industry investment, other institutions spent $19.1 billion on health research, an increase of about 5% from the previous year. Universities increased spending of institutional funds for research to $11.9 billion in 2010, a 6% increase. Philanthropic spending decreased slightly, while voluntary health groups increased investment in research by 15%, or $131 million, from the prior year.

According to funding projections in the report, the research investment landscape could worsen in 2013 and over the next decade. The scenario is different in other countries; as just one example, China has identified biotechnology as one of the seven "strategic and emerging (SEI) pillar" industries and has pledged to invest $308.5 billion in biotechnology over the next 5 years. Overall, the report provides analysis that outlines health research as one of the underpinnings of the U.S. economy and a key to improving the health of Americans.

Pending budget cuts will further jeopardize global leadership in research

WASHINGTON, DCOctober 25, 2012Biomedical and health research and development (R&D) spending from all sources declined by more than $4 billion or 3% between FY10 and FY11 according to Research!America's 2011 U.S. Investment in Health Research report. This represents the first drop in overall spending since Research!America began compiling the data in 2002.

The decline follows an uptick in research funding attributed to the American Recovery and Reinvestment Act (ARRA), which allocated $10.4 billion to the National Institutes of Health (NIH) over two fiscal years (2009-2010). The overall downward trend in R&D spending is coming at a time when other nations are ramping up their own investments in research, and meanwhile, pending across-the-board budget cuts (sequestration) could reduce federal biomedical and health research funding by 8%-10% or more.

Germany remembered the Holocaust's forgotten victims on Wednesday by opening a memorial in the heart of Berlin to the 500,000 ethnic Sinti and Roma murdered by the Nazis.

As the mournful strains of a solo violin sounded through the trees, political leaders and frail survivors approached a dark pool close to the German parliament building.

Its still water is intended to evoke tears for the dead but also, in reflecting the beholder, inspire new generations to protect minorities from hate.


"This memorial commemorates a group of victims who, for far too long, received far too little public recognition ? the many hundreds of thousands of Sinti and Roma who were persecuted by the Nazis as so-called gypsies," said German Chancellor Angela Merkel.?"The destiny of every single person murdered in this genocide is one of unspeakable suffering. Every single destiny, fills us, fills me, with sadness and shame."

The memorial was designed by the Israeli artist Dani Karavan, the BBC reported, and a chronology of the Nazis' extermination campaign appears next to the memorial. A fresh flower will be placed on the triangular surface at the center of the reflecting pool every day, the BBC said.

Discrimination against Sinti and Roma increased at alarming levels once Adolf Hitler took power in 1933. They were sent to forced labor camps and, from 1934, subjected to forced sterilization as a result of the Nazis' "racial purity" laws.

By the start of World War II, the Nazis' genocidal intent became clear as Sinti and Roma were deported to death camps, where they wore uniforms bearing a "Z" for "Zigeuner" (the German word for "gypsy").

The first time a German leader recognized Nazi persecution of the Roma on racist grounds was in 1982, more than 30 years after West Germany acknowledged the murder of 6 million Jews and began to pay compensation to Israel.

Discrimination today
German politicians and Roma leaders at the opening ceremony described the memorial as a reminder of the urgent need to protect minorities today.

Many of Europe's 12 million Roma face discrimination and social exclusion, often living in dire poverty.

"Half a million Sinti and Roma, men, women and children, were murdered during the Holocaust. Society has learned nothing, next to nothing from this, otherwise they would treat us differently," said Dutch Sinto survivor Zoni Weisz.

His voice faltered as he described how, as a seven-year-old, he watched his father, mother, sisters and brother being deported in a train to Auschwitz concentration camp.

Merkel stressed it was a German and European duty to protect Roma rights. After her speech, a heckler highlighted that Germany refuses to grant asylum to Roma from countries such as Serbia and Macedonia, where they face discrimination.

Romani Rose, leader of the Central Council of Sinti and Roma in Germany, also attended the ceremony.

"Opening the memorial sends an important message to society that anti-Roma sentiment is as unacceptable as anti-Semitism," he told AFP.

Montrose Swimming Pool Hall Closed Following Vandalism

Montrose swimming pool has been closed following vandalism to the pool over the weekend.

The pool was due to remain open until Sunday 11 November 2012, a few days before the town's new swimming pool and sports complex, at Marine Avenue, opens to the public on 17 November.

The weekend vandalism included the firing of missiles, stones and possibly a pool ball and resulted in two windows being smashed and glass falling into the water. The pool staff made several attempts to remove glass from the affected areas of the pool but following further inspections today by Angus Council officers, it was decided to close the pool, which would take several weeks and some considerable expense to drain, clean and refill. The swimming facilities will not be made available again at this site, but fitness and health suite facilities will continue to be available until 11 November 2012.

Council leisure spokesperson Councillor Jeanette Gaul said: "It is disappointing and disheartening that, as has sadly happened recently elsewhere in Angus, a tiny minority of anti-social individuals are spoiling the facilities which we provide, at great public expense and at times of severe budget cuts, for the benefit of the whole community.

"In effect, the children and adults of Montrose and further afield are being deprived of swimming facilities for several weeks due to the behaviour of a small number of people. I urge members of the public to be vigilant and to contact Tayside Police immediately if they have any information on this recent deplorable, disgraceful incident."

Endeavour rolls into new L.A. home at museum

The retired space shuttle Endeavour on Sunday entered the home stretch of its journey through the streets of Los Angeles to its retirement at a museum.

By 7:30 a.m. PT, the orbiter was still making its slow crawl toward Exposition Park, steered cautiously down Martin Luther King Blvd. as crowds gathered along both sidewalks. It was expected to reach final destination at the California Science Center later Sunday morning.

On Saturday, Endeavour rolled at a snail-like pace through narrow city streets, arriving five hours late at a key checkpoint. Enthusiasm remained high despite the slow pace, with an estimated 165,000 bystanders lining the streets to greet the spaceship.

Endeavour nosed out of Los Angeles International Airport before dawn on Friday for the 12-mile trip to its retirement home. Organizers had expected the shuttle to complete its journey on Saturday evening but it fell behind schedule crews had to make late adjustments to clear room for it.

Shuttle Endeavour is new L.A. 'star'

The shuttle, which has been a cause for cheers and expressions of awe from spectators watching it parade through the streets, will become a tourist attraction at the center. Endeavour was largely built in Southern California and was a workhorse of the U.S. space program, flying 25 missions.

Astronaut Michael Fincke, who went to space in Endeavour, said he and other astronauts on the shuttle's parade route felt the shuttle's road trip -- one unlike any voyage it has ever taken -- was special.

"We've seen our beautiful planet Earth from space, we've been weightless, we've been able to fly -- no special effects needed when you're in space," Fincke told the crowd outside a south Los Angeles shopping mall.

"And I tell you what, even though we've been in space we would not rather be anywhere else than where we are today," he said.

Organizers had planned to have the Endeavour arrive at the Baldwin Hills Crenshaw Plaza mall at 2 p.m. but instead it arrived after 7 p.m. about five hours behind schedule, said the organizers, a coalition that includes the Science Center and local authorities.

A huge crowd gathered outside the mall, where dancers and a high school marching band performed. It was a key checkpoint because the ship had to make a 90-degree turn to the east. The trip from the mall to the museum is about 4 miles.

Maintenance and tree trimming
The trip has been delayed in part due to maintenance needed for the massive, wheeled transporter carrying Endeavor and the need to trim some trees along the route, organizers said.

An estimated 100,000 spectators lined Martin Luther King Boulevard to watch the final, eastward leg of the journey through working-class south Los Angeles, a spokeswoman for the move's joint information center said.

Earlier in the day, about 65,000 people watched the shuttle head north along Crenshaw Boulevard, said Steve Ruda, a battalion chief for the Los Angeles Fire Department.

Thousands of spectators also watched earlier on Saturday when the shuttle stopped for a festival-like morning rally outside an arena in the nearby city of Inglewood.

Endeavour flew from 1992 to 2011 and was built to replace the Challenger, which exploded seconds into a 1986 launch that killed all seven crew members on board. Endeavour was taken out of service at the end of the shuttle program.

The shuttle is 122 feet long and 78 feet wide and stands 5 stories tall at the tail, which police said makes it the largest object ever to move through Los Angeles. Its combined weight with the transporter is 80 tons.

Organizers say only a few inches separate Endeavour's wings from structures along the route, and workers have felled 400 trees along curbs to clear a path. The science center will plant more than 1,000 trees to make up for their loss.

Some street lights, traffic signals, power poles and parking meters were temporarily removed.

The project to move Endeavour will cost more than $10 million, said Shell Amega, a science center spokeswoman. Charitable foundations and corporations have donated money and services for the move.

Endeavour has hop-scotched across the country from Cape Canaveral, Florida, on the back of a modified Boeing 747. It had been parked at the airport in Los Angeles since arriving on September 21 after a ceremonial piggyback flight around California.

The shuttle will be displayed in a temporary hangar-style metal structure to protect it from the elements. In 2017, a 200-foot-tall (61-meter) structure will open in which Endeavour will stand vertically, said Ken Phillips, aerospace curator at the California Science Center.

The other remaining shuttles also have found homes.

The Smithsonian in Washington has Discovery at its Steven F. Udvar-Hazy Center museum in Virginia. New York City has the prototype shuttle Enterprise at its Intrepid Sea, Air and Space Museum. And the Kennedy Space Center in Cape Canaveral has Atlantis, which the center will move to an on-site visitors complex next month.

Condom use drops when young women use hormonal contraceptives

ScienceDaily (Oct. 12, 2012) ? Young women who start using hormonal contraceptives for birth control often stop using condoms, but a new study in the Journal of Adolescent Health finds that if they later discontinue using hormonal contraceptives, they tend not to resume using condoms. This leaves them open to both unintended pregnancy and sexually transmitted infections (STIs).

The prospective year-long study included 1,194 sexually active women aged 15 to 24 visiting Planned Parenthood clinics and who were starting on contraceptive pills, patches, injections, or vaginal rings and not planning a pregnancy within the year. The researchers collected data on the young women's beliefs toward condom use and their knowledge of their partners' beliefs toward condoms.

At the beginning of the study, 36 percent of the young women used condoms consistently. Condom use dropped to 27 percent by 3 months later. Over the year, some women discontinued using hormonal contraception. More than half did not resume using condoms after they stopped using other contraception, according to Rachel Goldstein, M.D., of Stanford University School of Medicine and lead author of the study.

Using condoms together with other contraception (dual method use) decreases both the risk of pregnancy and sexually transmitted infections. The single largest predictor of using condoms and other contraception is the attitude of the woman's partner toward condoms, Goldstein noted. Women who said their partner thought condoms were "very important" or did not know how their partners felt about condom use were more likely to be dual method users than women who said their partner thought condoms were "not at all important."

"It appears that her partner's feelings may be more important than her perceived risk of a sexually transmitted infection or her own beliefs about dual method use," said Goldstein. This finding underscores the fact that a woman's decisions about contraception and preventing STIs may depend on factors over which she has little control. "Although a woman feels like she is at risk for an STI, she may not be able to advocate for herself and successfully negotiate condom use with her partner," Goldstein explained. Because a lapse in hormonal contraceptives may lead to unintended pregnancy, it may be better practice to advise young women to use longer-acting reversible contraceptives, such as implants or IUDs, and about the importance of condoms in preventing STIs, she recommended.

The study did not look at the use of hormonal implants or IUDs, also noted Mark Nichols, M.D., professor of obstetrics and gynecology at Oregon Health and Science University in Portland. IUDS, which protect against pregnancy for several years, are an increasingly popular and highly recommended choice for women who are starting contraception now. Furthermore, although the study concluded that more counseling was needed, it did not describe the kind of counseling the women received either when they started hormonal contraception or at later clinic visits, he added.

ANCHORAGE, Alaska (Reuters) - Alaska state employees can use private email accounts for public business but the messages must be preserved under public-records laws, the Alaska Supreme Court ruled on Friday in a case stemming from ex-Governor Sarah Palin's communications practices.

Palin is out of office but the ruling could affect her successor, Governor Sean Parnell, who is involved in his own controversy over public records. Critics have accused Parnell and his aides of using text messages rather than official emails to keep communications out of public view.

Electronic messages about state business are no different from paper communications under the Alaska Public Records Act, the Alaska Supreme Court said in its unanimous 16-page written decision that largely upheld a lower court ruling.

All paper or electronic messages regarding state business must be preserved and made available for public review, in accordance with state law, "and that duty cannot be extinguished by a public official's unreviewable decision simply not to preserve them," the court said.

A spokeswoman for Parnell said that his administration viewed the court's ruling as "favorable."

"Text messages are, by their nature, transitory, so they wouldn't be appropriate for preservation or constitute public records," Parnell's press secretary, Sharon Leighow, told Reuters in an emailed statement.

Friday's decision resulted from a lawsuit filed in 2008 by Andree McLeod, an Anchorage activist who criticized Palin, the Republican nominee for vice president that year, for using private Yahoo accounts to communicate state business.

She and others contended that Palin used private accounts to evade state public-records rules. McLeod also argued that no state business should be conducted on private email accounts.

Open-records requests filed by McLeod and several journalists resulted in release of thousands of pages of Palin emails during her term as governor, many of them from her private accounts. The emails were released in 2011 and earlier this year, long after Palin resigned her post in 2009.

Friday's ruling, though it partially affirmed the lower-court ruling protecting the use of private emails, is a victory for McLeod, her attorney said.

"The Alaska Supreme Court's decision this morning demonstrates the importance of what citizen activists such as Andree McLeod can accomplish to advance the interests of all Alaskans in ensuring that all state employees, from the governor on down, conduct official state business in a manner that at all times and through all means of communication serves the public interest," attorney Don Mitchell said in a statement.

An August 27 opinion by the state legislature's legal adviser concluded that text messages are subject to state public records laws. Parnell's spokeswoman said at the time that the governor disagrees and considers text messages to be "transitory in nature" and difficult to preserve.

A representative for Palin could not immediately be reached for comment.

Fly genomes show natural selection and return to Africa

ScienceDaily (Oct. 11, 2012) ? When ancestral humans walked out of Africa tens of thousands of years ago, Drosophila melanogaster fruit flies came along with them. Now the fruit flies, widely used for genetics research, are returning to Africa and establishing new populations alongside flies that never left -- offering new insights into the forces that shape genetic variation.

That's one of the findings from two new papers published this month by researchers at the University of California, Davis, and their colleagues that describe the genomes of almost 200 strains of the tiny flies.

The work reveals strong evidence of pervasive natural selection throughout the D. melanogaster genome, said Charles Langley, professor of genetics in the Department of Evolution and Ecology at UC Davis and an author on both papers. That is in striking contrast with what is known of the human genome, which shows comparatively little evidence of adaptation over the last 100,000 years.

The overall aim of the research is to better understand the forces that shape genetic variation, Langley said. Human geneticists are working to sequence a thousand human genomes to get at the same issue. The knowledge from studying fly genetics has and will help in that effort, he said.

"Most of the theory and statistical methods in human genetics were originally motivated by studies of flies, because they're easier and faster to work with," Langley said. "Model organisms like these play a critical role in developing tools and ideas."

The first paper, published Oct. 1 in the journal Genetics, reports the genomes of 37 strains of Drosophila collected in Raleigh, N.C., and six strains from the sub-Saharan nation of Malawi. The other paper, to be published in PLoS Genetics and currently available online as a preprint, describes the genomes of 139 fly strains covering 22 African and one European population.

Drosophila melanogaster, like humans, originated in Africa, and that's where they show the most genetic diversity -- just as humans do. The flies are thought to have appeared in Europe about 50,000 years ago, along with modern humans. On the way, both humans and flies squeezed through genetic bottlenecks that reduced the diversity in the population. Over generations, different fly strains evolved quite specialized niches -- such as a fondness for colonizing breweries, for example.

But the African fly genomes show that just in the past couple of decades, flies similar to those found in Europe or the U.S. have established populations back in Africa, often in new environments such as urban and industrial developments. For example, where modern breweries and bottled beer have displaced traditional African brewing, "Europeanized" brewery flies have followed.

The spread of those "European" genes is more rapid than if it were occurring by random processes, the researchers write.

"It may be that urbanization and development favor the more 'European' flies," Langley said.

UC Davis is internationally recognized for research in evolution, ecology and genetics. Campus researchers have collaborated on efforts to sequence the genomes of a wide variety of plants and animals, including wheat, rice, cucumber, horses, cats and chickens.

Langley's UC Davis-affiliated co-authors on the Genetics paper are researchers Kristian Stevens and Charis Cardeno, graduate students Yuh Chwen Lee, Sasha Langley, Charlyn Suarez and Russell Corbett-Detig, Professor David Begun, and postdoctoral researcher John Pool, all in the Department of Evolution and Ecology. Other co-authors include researchers at Indiana University, Bloomington; Lawrence Berkeley National Laboratory; University of Florida, Gainesville; UCSF; Rutgers University; Academica Sinica, Taiwan; and UC Berkeley.

Pool, now at the University of Wisconsin-Madison, is first author on the upcoming PLoS Genetics paper, with Langley, Corbett-Detig, Stevens, Cardeno, Begun, researchers Marc Crepeau and Perot Saelao from UC Davis, along with researchers from UW Madison and the University of Munich, Germany.

Romney criticizes Biden on consulate attack

RICHMOND, Va. (AP) ? Broadening his attack on administration foreign policy, Mitt Romney accused Vice President Joe Biden on Friday of "doubling down on denial" in a dispute over security at a diplomatic post in Libya that was overrun by terrorists who killed the U.S. ambassador and three other Americans.

"The vice president directly contradicted the sworn testimony of State Department officials," the Republican presidential candidate said, eager to stoke a controversy that has flared periodically since the attack on Sept. 11 "... American citizens have a right to know just what's going on. And we're going to find out."

President Barack Obama had no campaign appearances during the day, leaving it to White House press secretary Jay Carney to defend Biden's assertion in a campaign debate Thursday night that "we weren't told" of an official request for more security at the site.

The spokesman rejected Romney's claim of a contradiction. Biden "was speaking directly for himself and for the president. He meant the White House," Carney said.

With his accusation, Romney once again pushed foreign policy to the forefront of a campaign dominated for more than a year by the economy, which has been painfully slow to recover from the worst recession in more than a half century.

The Republican challenger was campaigning across a pair of battleground states during the day, first in Virginia, which has 13 electoral votes, and then in Ohio, which has 18 electoral votes and where running mate Paul Ryan joined him. It takes 270 electoral votes to win the White House.

Biden was in Wisconsin, Ryan's home state, and one where polls give Obama a narrow lead despite a debate performance last week that was so poor it fueled a Republican comeback nationally and sent shudders through the ranks of Democratic partisans.

More than a week later, officials in both parties describe a race that has largely returned to the competitive situation in effect last summer, before the national political conventions and the emergence of a videotape in which Romney spoke dismissively of nearly half the country propelled the president to significant gains in the polls.

Now, many of the same surveys show a very tight race nationally and in most of the competitive states, although the president holds a small lead in public and private surveys in Ohio and Wisconsin.

Still struggling to blunt or reverse Romney's rise in the polls, Obama's campaign launched two new ads in several of the contested states. One shows the Republican being asked in a "60 Minutes" interview if it's fair that he paid federal tax of about 14 percent last year on income of $20 million, while a $50,000 wage-earner paid a higher rate. "I think it's the right way to encourage economic growth," he says, and the narrator adds: "Lower tax rates for him than us. Is that the way to grow America?"

The second commercial appears aimed at recent comments Romney made suggesting he might not make opposition to abortion a priority. "Maybe you're wondering what to believe about Mitt Romney," it says, then shows him pledging to eliminate federal funding for Planned Parenthood.

With control of the Senate and all 435 House seats at stake along with the White House, outside groups that spent months stockpiling money were now in a race to spend it.

American Crossroads, a group backed by former White House strategist Karl Rove, announced this week it was spending $7.4 million in the presidential race, while an allied organization, Crossroads GPS, put down $4 million to help Republicans in five Senate races and another $8.1 million for 11 House campaigns ? a total of nearly $20 million.

Some candidates seemed to be showing signs of campaign fatigue.

In a California House race between two Democrats, Rep. Brad Sherman seized the shoulder of Rep. Howard Berman during a debate, yanked him toward his chest and shouted, "You want to get into this?" The two men stood nose to nose before a sheriff's deputy moved between them.

"I should not have done that," conceded Sherman, 57, on Friday.

Said his 71-year-old rival: "It was like in the eighth grade, 'You want to go over to the park on the corner and fight this out?'"

The two Democrats are pitted against each other because California advances the top two vote-getters in a primary to the general election, regardless of their party.

In the presidential race, Romney began the campaign week with a speech that criticized the Obama administration for showing a lack of leadership around the globe, particularly in the Middle East.

And he chose to end it with a direct challenge to Biden's candor about the attack on the consulate in Benghazi, Libya.

"When the vice president of the United States directly contradicts the testimony, sworn testimony of State Department officials, American citizens have a right to know just what's going on," he said, referring to a hearing earlier in the week in a Republican-controlled House committee.

One official testified before the panel that he had been criticized for seeking additional security at the facility. A second said she personally had turned down requests for more protection at the facility in Benghazi.

Carney said, that despite Romney's allegation, there was no contradiction between what Biden said and what the congressional committee had been told.

"Requests for individual personnel at the thousands of facilities ... are not adjudicated at the White House," the spokesman said. "They are decided at the State Department."

Biden, campaigning in LaCrosse, Wis., did not mention Libya on the day after the debate. Instead, he mocked Ryan for having said on Thursday night that a House budget proposal that he authored would not lead to drastic spending cuts in Medicare, education and other areas.

"Congressman Ryan saying his budget does not have spending cuts is like Gov. Romney standing in an unemployment line and saying, 'I didn't outsource you job, I offshored it," he said, referring to a distinction Republicans sought to draw earlier in the campaign.

The controversy over Libya flared as both Romney and Obama looked ahead to their second debate, set for next Tuesday in Hempstead, N.Y.

After being accused by some Democrats of failing to prepare adequately for last week's encounter, Obama arranged for several days of rehearsals in Williamsburg, Va.

Romney was flying home to Massachusetts on Saturday so he, too, could get ready for an event likely to be watched by a television audience measured in the tens of millions.

The two men will hold their third and final debate on Oct. 22 in Boca Raton, Fla.
